I go With Ad-Aware SE and SPYbot Seek and destroy

I use both in tandem, and does the job. Plus if they can't get rid of sometihng, I then know where it is, and I'll manually and forcefully remove it.

I use IE6 and Firefox (IE for normal browsing, and firefox for crossbrowser testing for sites and a backup browser)
